Previous studies on in vitro rubber biosynthesis (RB) suggested that the washed rubber particles (WRP) surface could catalyze the formation of new rubber molecules from allylic initiators. However, this need to be reconsidered as shown by our earlier report that new rubber formation can occur at certain site other than WRP surface*. In this further systemic study, initiation of new rubber molecules was unequivocally demonstrated in washed bottom fraction particles. (WRP), sediment membrane bound particles of the centrifuged fresh Hevea latex. The washed bottom fraction particles membrane (WRM) prepared from WBP was shown to exhibit much higher RB level. The membrane function of WBM for RB activity was stimulated by anionic detergent (SDS), but nonionic detergents (Triton X-100 and Tween 20) were without any effect. Heat preincubation treatment of WBM led to a much higher RB stimulation than the SDS effect. The heat pretreated WBM activity exhibited much more pronounced RB stimulation when treated in the presence of SDS detergent, indicating synergistic effect. Heat activation of WBM was at a much lower temperature with SDS and still with higher RB stimulation. Synergistic stimulation on the membrane RB activity of WBM could open up a novel bio-reactor for specialty natural rubber synthesis. *Rubber formation by fresh bottom fraction of Hevea latex (Phytochemistry 45, 269.)
